Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Solid material is generally held at -20 °C or colder, while reconstituted solutions are kept at 2-8 °C and used within a short window. Buffers that maintain a slightly acidic to neutral pH tend to improve short-term peptide stability. Repeated warming and cooling of stock solutions promotes aggregation and should be avoided. Container closures should remain intact, since adsorption to some plastics can reduce the amount of peptide in solution.
Identity and purity are established with reversed-phase high-performance liquid chromatography and mass spectrometry. Chromatographic profiles reveal related impurities, truncated sequences, and oxidation products, while mass measurement confirms the expected molecular mass. Purity values for research material are typically reported as a percentage by peak area. Reference standards help calibrate retention behavior across instruments. Independent laboratories emphasize method suitability because results depend heavily on column chemistry, gradient, and detection wavelength. Batch-to-batch comparison relies on the same validated method.

Mechanistic proposals link each receptor to a different physiological role. Activation of the glucose-dependent insulinotropic polypeptide and glucagon-like peptide-1 receptors is associated with reduced appetite, slower gastric emptying, and glucose-dependent insulin release. Glucagon receptor signaling, by contrast, is associated with increased energy expenditure and altered lipid handling, though it can also raise blood glucose. The design intent is to balance these contributions so that weight reduction is enhanced without unacceptable glycemic trade-offs. How well that balance holds across individuals is not fully resolved.
Published information places retatrutide in clinical development rather than on the market as an approved therapy. Early-stage and mid-stage trials have examined tolerability and changes in body weight, and larger studies continue to report results over time. Open questions include the durability of effects after treatment stops, the composition of weight lost, and cardiovascular outcomes over long periods. Statements about definitive benefit should therefore be treated as provisional. Regulatory status varies by jurisdiction and changes as applications are reviewed.
Retatrutide is an investigational synthetic peptide designed to activate three distinct receptor systems within a single molecule. Its pharmacological profile combines activity at the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor. This arrangement places it within a broader class of agents often described as multi-agonists, which contrast with compounds that engage one or two targets. Research interest centers on whether simultaneous signaling produces effects that single-receptor agonists cannot achieve alone. A single molecular entity also simplifies manufacturing and delivery logistics compared with combining separate agents.
Troglitazone is an antidiabetic and anti-inflammatory drug, and a member of the drug class of the thiazolidinediones. It was prescribed for people with diabetes mellitus type 2. It was patented in 1983 and approved for medical use in 1997. It was subsequently withdrawn.
=== Brazil === The R2 Combat Operation Ration (Portuguese: Ração Operacional de Combate – R2) is the current field & combat ration for the Brazilian Army. It is based on the earlier, but similar, Alternative Combat Ration for 24 Hours (Portuguese: Ração Alternativa de Combate, 24 horas) developed by the Brazilian Navy for use by Naval Infantry units. It contains the food and supplemental items needed by 1 soldier for 24 hours. It is to be used in situations where no other type of ration is available. All foods are packed inside 4-ply plastic and aluminum polylaminate retort pouches and are ready to eat without further preparation. The ration is packed inside a heavy-duty (.25 mm thick) matte green or olive drab polyethylene bag measuring 300 mm wide by 400 mm long. It is printed with the logo of the Brazilian Army, the name of the ration, and menu information. Inside are 5 thinner (.10 mm) semi-transparent plastic bags, one for each meal and one for the accessories. Each bag is printed with meal information and contents.
Anti-Ro and anti-La antibodies, also known as SS-A and SS-B, respectively, are commonly found in primary Sjögren's syndrome, an autoimmune disorder that affects the exocrine glands. The presence of both antibodies is found in 30–60% of Sjögren's syndrome, anti-Ro antibodies alone are found in 50–70% of Sjögren's syndrome and 30% of SLE with cutaneous involvement, and anti-La antibodies are rarely found in isolation. Anti-La antibodies are also found in SLE; however, Sjögren's syndrome is normally also present. Anti-Ro antibodies are also found less frequently in other disorders including autoimmune liver diseases, coeliac disease, autoimmune rheumatic diseases, cardiac neonatal lupus erythematosus and polymyositis. During pregnancy, anti-Ro antibodies can cross the placenta and cause heart block and neonatal lupus in babies. In Sjögren's syndrome, anti-Ro and anti-La antibodies correlate with early onset, increased disease duration, parotid gland enlargement, disease outside the glands and infiltration of glands by lymphocytes. Anti-Ro antibodies are specific to components of the Ro-RNP complex, comprising 45kDa, 52kDa, 54kDa and 60kDa proteins and RNA. The 60kDa DNA/RNA binding protein and 52kDa T-cell regulatory protein are the best characterised antigens of anti-Ro antibodies. Collectively, these proteins are part of a ribonucleoprotein (RNP) complex that associate with the human Y RNAs, hY1-hY5. The La antigen is a 48kDa transcription termination factor of RNA polymerase III, which associates with the Ro-RNP complex.
The liver is a vital organ and supports almost every other organ in the body. Severe or end-stage liver failure has dire consequences for the body's overall health and quality of life. Visible signs of liver disease include jaundice and ascites. Hepatomegaly refers to an enlarged liver and may be caused by several underlying diseases. It can be palpated in an abdominal exam. Scarring (fibrosis) of the liver from any cause can lead to cirrhosis. Cirrhosis increases the resistance to blood flow in the liver, and can result in portal hypertension. Congested anastomoses between the portal venous system and the systemic circulation, can be a subsequent condition. The most common chronic liver disease is nonalcoholic fatty liver disease, which affects an estimated one-third of the world population. Hepatitis is a common condition of inflammation of the liver. The cause is usually viral, and the most common of these infections are hepatitis A, B, C, D, and E. Some of these infections are transmitted sexually or through non-sterile needles. Inflammation can also be caused by other viruses in the family Herpesviridae such as the herpes simplex virus. Chronic (rather than acute) infection with hepatitis B virus or hepatitis C virus is the main cause of liver cancer. Globally, about 248 million individuals are chronically infected with hepatitis B (with 843,724 in the U.S.), and 142 million are chronically infected with hepatitis C (with 2.7 million in the U.S.).

== Hormones == Estrogens cause the hips to widen and become rounded during puberty in females, and androgens cause the shoulders to broaden in males. Estrogens mediate epiphyseal closure in both males and females. Other hormones implicated in control of bone growth include thyroid hormone, parathyroid hormone, calcitonin, glucocorticoids such as cortisol, and vitamin D (calcitriol). According to menoPAUSE, a blog from University of Rochester, estrogen causes females to have their fat distributed in their breasts, thighs, and along their pelvic area, implying that the fat can be used as an energy source for future pregnancies. For men, androgens (such as testosterone) increases male's muscle-to-fat ratio.Woods J. "What does Estrogen Have to Do with Belly Fat?".
== Composition == Compared to their prokaryotic homologs, many of the eukaryotic ribosomal proteins are enlarged by insertions or extensions to the conserved core. Furthermore, several additional proteins are found in the small and large subunits of eukaryotic ribosomes, which do not have prokaryotic homologs. The 40S subunit contains a 18S ribosomal RNA (abbreviated 18S rRNA), which is homologous to the prokaryotic 16S rRNA. The 60S subunit contains a 28S rRNA that is homologous to the prokaryotic 23S ribosomal RNA. In addition, it contains a 5.8S rRNA that corresponds to the 5' end of the 23S rRNA, and a short 5S rRNA. Both 18S and 28S have multiple insertions to the core rRNA fold of their prokaryotic counterparts, which are called expansion segments. For a detailed list of proteins, including archaeal and bacterial homologs please refer to the separate articles on the 40S and 60S subunits. Recent research suggests heterogeneity in the ribosomal composition, i.e., that the stoichiometry among core ribosomal proteins in wild-type yeast cells and embryonic stem cells depends both on the growth conditions and on the number of ribosomes bound per mRNA.
When the British missed capturing President Paul Kruger, as he escaped Pretoria during its fall in June 1900, a Melbourne Punch cartoon depicted how the War could be won, using the Kelly Gang. The convictions and executions of two Australian lieutenants, Harry Harbord Morant and Peter Handcock in 1902, and the imprisonment of a third, George Witton, had minimal impact on the Australian public at the time. The controversial court-martial saw the three convicted of executing prisoners under their authority. After the war though, Australians joined an empire-wide campaign that saw Witton released from jail. Much later, some Australians came to see the execution of Morant and Handcock as instances of wrongfully executed Australians, as illustrated in the 1980 Australian film Breaker Morant. Up to 50 Aboriginal Australians served in the Boer War as trackers. Such is the lack of information available it is even uncertain as to whether they returned to Australia after the war. When the Australian contingents returned the trackers may not have been allowed back to Australia due to the White Australia Policy.

=== CSIR India === In 2009, Ayyadurai was hired by India's Council of Scientific and Industrial Research (CSIR), India's largest science agency, by its director general, Samir K. Brahmachari. CSIR was mandated to create a new company, CSIR Tech, that would establish businesses using the research conducted by the country's many publicly owned laboratories. Ayyadurai reported that he had spent months trying to create a business plan for CSIR Tech, but received no response from Brahmachari. Ayyadurai then distributed a draft plan, which was not authorized by CSIR, to the agency's scientists that requested feedback and criticized management. His job offer was subsequently withdrawn five months after the position was offered. Brahmachari said that "the offer was withdrawn as [Ayyadurai] did not accept the terms and conditions and demanded unreasonable compensation." In its report, The New York Times said that "going public with such accusations is highly unusual. Mr. Ayyadurai circulated his paper not just to the agency's scientists but to journalists, and wrote about his situation to Prime Minister Manmohan Singh." In that letter, Ayyadurai said his report was intended to explore institutional barriers to CSIR's entrepreneurial mandate. He said that CSIR scientists reported that "they work in a medieval, feudal environment" that required a "major overhaul". The letter was co-authored by a colleague, Deepak Sardana.
A 2016 WHO report found that formaldehyde from second-hand vapor was around 20% greater compared to background air levels. Normal usage of e-cigarettes generates very low levels of formaldehyde. Different power settings reached significant differences in the amount of formaldehyde in the e-cigarette vapor across different devices. Later-generation e-cigarette devices can create greater amounts of carcinogens. Some later-generation e-cigarettes let users increase the volume of vapor by adjusting the battery output voltage. Depending on the heating temperature, the carcinogens in the e-cigarette vapor may surpass the levels of cigarette smoke. E-cigarettes devices using higher voltage batteries can produce carcinogens including formaldehyde at levels comparable to cigarette smoke. The later-generation and "tank-style" devices with higher voltages (5.0 V) could produce formaldehyde at comparable or greater levels than in cigarette smoke. A 2015 study hypothesized from the data that at high voltage (5.0 V), a user, "vaping at a rate of 3 mL/day, would inhale 14.4 ± 3.3 mg of formaldehyde per day in formaldehyde-releasing agents." The 2015 study used a puffing machine showed that a third-generation e-cigarette turned on to the maximum setting would create levels of formaldehyde between five and 15 times greater than with cigarette smoke.
== Function == Following binding of GnRH, the GnRHR associates with G-proteins that activate a phosphatidylinositol (PtdIns)-calcium second messenger system. Activation of the GnRHR ultimately causes the release of follicle stimulating hormone (FSH) and luteinizing hormone (LH).
== Military campaigns in Syria and Jazira == Ubayd Allah fought for Marwan and his tribal allies against the Qaysi tribes led by al-Dahhak ibn Qays al-Fihri, the governor of Damascus, at the Battle of Marj Rahit in August 684. The Qays were routed and al-Dahhak killed. Ubayd Allah was put in command of Marwan's army which, during Marj Rahit, consisted of 6,000 men from a handful of loyalist tribes. According to Kennedy, Ubayd Allah "clearly intended to rebuild the Syrian army which had served Mu'awiya and Yazid I so well". In the aftermath of Marj Rahit, Ubayd Allah oversaw campaigns against rebel Qaysi tribes for Marwan and his son and successor Abd al-Malik (r. 685–705) in the Jazira. However, Marwan's forces were too little to assert Umayyad rule throughout the caliphate. Thus, Ubayd Allah expanded recruitment to include various Qaysi tribes. He placed Husayn ibn Numayr al-Sakuni of Kindah as his second-in-command, and Shurahbil ibn Dhi'l Kala' of Himyar, Adham ibn Muhriz of Bahila, Rabi'a ibn al-Mukhariq of Banu Ghani and Jabala ibn Abd Allah of Khath'am as deputy commanders. Other than Husayn ibn Numayr, all of the commanders were either Qaysi or had earlier supported al-Dahhak against Marwan. In January 685, as Ubayd Allah was in Manbij preparing for the Umayyad reconquest of Iraq, Husayn ibn Numayr defeated the pro-Alid Penitents at the Battle of Ayn al-Warda. Ubayd Allah had been promised by Marwan the governorship over all of the lands he could conquer from the Alids and Ibn al-Zubayr, and he may have been sanctioned to plunder Kufa.
=== Biosynthetic pathway === The metabolic pathway to produce ergothioneine starts with the methylation of histidine to produce histidine betaine (hercynine). The sulfur atom is then incorporated from cysteine. The biosynthetic genes of ergothioneine have been described in detail for Mycobacterium smegmatis, Neurospora crassa, Schizosaccharomyces pombe (with homologues in Aspergillus, a genus important in food fermentation), and Caldithrix abyssi. This pathway has recently also been discovered in plants. Different groups of organisms use different approaches to sulfur-addition. Aerobic bacteria and fungi use an O2-dependent reaction that is catalyzed by a mononuclear non-heme iron enzyme, with cysteine or γ-glutamylcysteine as the sulfur source. Green sulfur bacteria and some archaea use a rhodanese-like sulfur transferase to perform oxidative polar substitution. Caldithrix uses a metallopterin-dependent bifunctional enzyme that combines an N-terminal domain similar to a tungsten-dependent acetylene hydratase and a C-terminal cysteine desulfurase domain. Homologs of the Caldithrix system are found in anaerobic bacteria and some archaea.

It is a synthetic peptide triagonist that engages GIP, GLP-1, and glucagon receptors. Investigational compounds in this class are studied for metabolic conditions rather than for a single organ system.
No regulatory agency has approved it for any indication. It remains an investigational product studied within controlled clinical trial programs.
A dual agonist engages two receptor targets, typically GIP and GLP-1. A triagonist adds glucagon receptor activity, which is intended to influence energy expenditure alongside appetite and glucose handling.
Solid powder is held frozen at -20 °C or below in a desiccated container. Reconstituted solutions are refrigerated and used within a limited period.
